INFLUENCE OF TEMPERATURE ON THE RESPONSES TO CALCIUM OF THE RAT TAIL ARTERY, RABBIT EAR ARTERY AND CAR PAW ARTERY

Smooth muscle tone and sensitivity in several vascular beds are known to change with temperature. While there is considerable evidence for radical changes in vascular smooth muscle responses to temperature changes, the
influence of desensitization and sensitivity to calcium and Bay K 8644 or nifedipine under these conditions are unknown and this study aimed at investigating these in rat tail artery, rabbit ear artery and cat paw artery.

GLUCOSE CONCENTRATION AND CONTRACTILE RESPONSES IN THE RAT TAIL ARTERY

The effect of half, double, and quadriple concentrations of the normal (2gm/litre or 10mM) concentration of glucose in the Kreb’s physiological saline were tested on vascular tissues activated by 3M noradrenalline or 100mM potassium chloride olution. The smooth muscle tissues were from isolated tail artery which were perfused with and bathed with calcium buffered physiological saline and with which six consecutive calcium concentration response curves were constructed  for each glucose concentration.

EFFECTS OF OXYGEN ON THE RAT TAIL ARTERY

The general procedure for maintaining isolated tissues in vitro is to equilibrate at atmospheric pressure, with a gas mixture of high oxygen content, such as 100% or where CO2 / bicarbonate buffering is used, 95% O2 is employed. But in a classical resistance vessel, such as the rat tail artery, little attention has been paid to the difference between normoxia of 16% and hyperoxia of 90% O2 .

EFFECTS OF BAY K 8644 AND NIFEDIPINE ON THE CALCIUM CONCENTRATION-RESPONSE CURVE AND ON DESENSITIZATION OF THE RAT TAIL ARTERY

It has been shown that desensitization of the responses of vascular smooth muscle could occur if the physiological saline composition is altered. The extent to which Bay K 8644 and nifedipine affect the calcium responses of a vascular
smooth muscle and their effects on desensitized tissue responses are yet to be quantified and this is what this study aims at doing on calcium-mediated responses in the rat tail artery and on the desensitization of the responses of the

EFFECT OF CALCIUM BUFFERS (EGTA AND NTA) ON THE RESPONSES OF THE RAT TAIL ARTERY

For experiments on smooth muscles into the effects of adrenaline, noradrenaline and various adrenoceptor agonists and antagonists, a range of extracullular calcium [Ca2+] needed to be established; thus necessitating the use of suitable calcium buffers in the bathing media. The extent to which NTA/EGTA calcium buffers affect the responsiveness of the rat tail artery has not yet been established, hence this study.

DESENSITIZATION OF RESPONSES TO NORADRENALINE AND POTASSIUM CHLORIDE IN THE RAT TAIL ARTERY

Previous investigators could not account for the factors causing the initial decline between the first and subsequent responses. They rather had neglected the first  few responses or the first concentration response curve (CRC) in the name of allowing the tissue to stabilize. The second or even the third CRC is often taken to represent the control curve for the analysis of drugs.
